Gene Variant Descriptions PTEN N276S lies within the C2 tensin-type domain of the Pten protein (UniProt.org). N276S demonstrates phosphatase activity similar to wild-type Pten in yeast (PMID: 21828076), but reduced Akt phosphorylation in culture and reduced expression in culture and in patient samples (PMID: 36591942), is unable to localize to the nucleus (PMID: 29373119), and leads to reduced Pten protein stability (PMID: 25527629, PMID: 32350270), and fails to suppress Akt phosphorylation in cell culture, and delays developmental growth in flies (PMID: 32350270).
