Y65C confers a loss of function to the Pten protein as demonstrated by reduced ATP-binding, nuclear accumulation (PMID: 20926450), increased cell proliferation, decreased apoptosis, and is transforming in culture (PMID: 19457929), and leads to reduced Pten protein stability, fails to suppress Akt phosphorylation in cell culture, and leads to impaired developmental growth in flies (PMID: 32350270). |
