Gene Variant Descriptions PTEN Q399* results in a premature truncation of the Pten protein at amino acid 399 of 403 (UniProt.org). Q399* results in reduced ability to inhibit PDGF-induced membrane ruffling and proliferation in PTEN-null cells (PMID: 10698713), fails to rescue epithelial morphogenesis in culture (PMID: 23085752), abrogates fibroblast conditioned medium-induced PI3K activation in culture (PMID: 35719951), but maintains similar levels of phosphatase activity to wild-type Pten in cell culture (PMID: 10698713), and therefore, its effect on Pten protein function is unknown.
